Abstract: The penalized D-optimal design is introduced in Dragalin and Fedorov (2006). We extend this method to the Bayesian realm for bivariate models of toxicity and efficacy. We compare our method to the Bayesian adaptive design of Thall and Cook (2004), which does not involve formal optimality criteria or efficiency considerations. The penalized optimal design methods tend to be more favorable in terms of consistency across simulations and information gain.
